The Effects of Lipid a on Gamma-irradiated Human Peripheral Blood Lymphocytes in Vitro

چکیده
The modulatory effects of lipid A (diphosphoryl lipid A (DLA) and monophosphoryl lipid A (MLA)) on apoptosis induction and DNA structure damage (singleand double-strand breaks (SSBs and DSBs, respectively)) in peripheral human blood lymphocytes are studied for Co gamma irradiation. It is shown that in the presence of these agents the number of apoptotic cells increases compared with the irradiated control samples. The effect is most strongly pronounced for DLA. In its presence, a signiˇcant increase is observed in the number of radiation-induced DNA SSBs and DSBs. Possible mechanisms are discussed of the modifying in uence of the used agents on radiation-induced cell reactions.

Assessment of the radioprotective effects of amifostine on human lymphocytes irradiated in vitro by gamma-rays using cytokinesis-blocked micronucleus assay
Background: A radioprotective effect of amifostine as well as its ability to modulate the level of spontaneous and gamma-irradiation-induced genetic changes on human peripheral blood lymphocytes has been investigated . Amifostine, known as a potent radical scavenger, has been introduced as the most effective radioprotector, yet it is not completely approved for the clinical use. However, furt...
متن کاملRadio-adaptive response of peripheral blood lymphocytes following bystander effects induced by preirradiated CHO-K1 cells using the micronucleus assay
Background: Radio-adaptive response and bystander effects are known phenomena occurring in cells following exposure to ionizing radiation (IR). In this study we examined possible radio-adaptation of lymphocytes following bystander effects induced by CHO-K1 cells. Materials and Methods: Whole blood and CHO-K1 cells were cultured in RPMI-1640 complete medium. Cells were separately irradiated with...
متن کاملIn vitro radioprotective effects of histamine H2 receptor antagonist against gamma-rays induced chromosomal aberrations in human lymphocytes.
Background: Radioprotective capability of histamine H2 receptor antagonists have been shown in several in vivo studies mainly using animal models. However, to verify the effectiveness of these agents in clinical applications, studies should be performed on human cells. In the present study radioprotective properties of these agents was examined in vitro on human lymphocytes using metaphase anal...

Background: To evaluate the effects of hyperthermia (HT) on the frequency of chromosomal aberrations induced by a low dose of neutron or γ-rays in human peripheral blood lymphocytes. Materials and Methods: Blood samples were exposed to HT (41.5°C for 30 and 60min, 43°C for 15 and 30min), 10 cGy neutron or γ-rays, HT + neutron/γ, and neutron/γ + HT. After standard cell culture, harvestin...
متن کاملEffects of 35 Hz, 2 mT Magnetic Field on Peripheral Blood Lymphocytes of Human In Vitro and Rat In Vivo
In this research we have studied the effects of extremely low frequency magnetic field on the peripheral blood lymphocytes chromosomes of human in vitro and rat in vivo. By this means, we used 60 blood samples of 10 men as well as 40 rats. Of 60 human blood samples ,40 samples (in two group of 20) were selected as test groups and placed into the 2 mT and 35 Hz magnetic field. The blood samples,...
